좌골신경의 압박성 손상을 입은 쥐에게 Berberine을 투여했을 때의 항이질통 효과

Abstract
The objective of this study was to investigate whether berberine could ameliorate allodynia induced by chronic constriction injury (CCI) of the sciatic nerve in rats. A small vertical incision was made along the thigh. The muscles of the posterior thigh (including the hamstring muscles) were split and the left sciatic nerve exposed at the mid-thigh level proximal to the sciatic trifurcation. Four chromic gut ligatures (4/0) were tied loosely around the nerve. After inducement of CCI, significant increase in the number of paw lifts at cold plate test (cold allodynia) and decrease of paw withdrawal threshold at von Frey hair stimulation test (mechanical allodynia) were observed. However, these cold and mechanical allodynia were markedly alleviated by berberine administration in a dose-dependent manner. Sciatic nerve myeloperoxidase and malondialdehyde activities were also attenuated by berberine administration. Continuous injection for maintenance therapy induced no development of tolerance. The anti-allodynic effect of berberine 20 mg/kg was comparable to that of amitriptyline 10 mg/kg. This study demonstrated that berberine can mitigate allodynia induced by CCI, a neuropathic pain model, and it had relevance to anti-inflammatory and anti-oxidative effects.
본 연구는 쥐에서 좌골신경에 압박성 손상을 주었을 때 berberine이 이질통을 개선할 수 있는지를 조사하기 위한 것이다. 좌골신경 결찰을 시행하고 난 후 cold plate test에서 발 드는 회수(한랭 이질통)가 명백히 증가하는 것을 보였고, von Frey hair stimulation test에서 회피 역치(기계적 이질통)가 감소하는 것이 관찰되었다. 그러나 이러한 한랭, 기계적 이질통은 용량 비례적으로 berberine 투여로 인해 두드러지게 완화되는 것으로 나타났다. 또한 좌골신경의 myeloperoxidase와 malondialdehyde 수치 또한 berberine 투여로 인해 감소되는 것을 보였고, 지속적 주입에도 내성이 발생하지 않았다. Berberine(20 mg/kg)의 항이질통 효과는 amitriptyline(10 mg/kg)의 효과와 비슷하게 나타났다. 본 연구에서 berberine은 만성 좌골신경 압박성 신경병증 모델인 신경 압박 손상으로 인한 이질통을 완화시킬 수 있는 것으로 나타났고, 항염증 효과, 항산화 효과와도 관련이 있는 것으로 사료된다.
