Abstract
동맥경화증은 여러 가지 요인들에 의해 발생되는 만성적인 혈관의 염증성 질병이며, 그 중에서 활성산소 생성증가와 산화형 저밀도 지단백이 동맥경화증 유발과 진행에 중요한 인자로 작용한다. Lysophosphatidylcholine(LPC)는 산화형 저밀도 지단백의 주성분으로 동맥경화증을 일으키는데 깊이 관여한다. 이번 연구에서는 인간 제대정맥 내피세포(human umbilical vein endothelial cells, HUVECs)에 대한 LPC의 영향을 파악하고, 과일과 커피에 다량 포함되어 있는 폴리페놀 성분인 chlorogenic acid의 보호효과와 그 작용기전을 규명하고자 하였다. HUVECs에 대한 LPC의 영향과 chlorogenic acid의 효과를 보기 위해 세포내 Ca2+ 농도, 세포생존율 및 활성산소 생성을 측정하였다. Ca2+ 통로의 활성 변화는 역전사중합효소연쇄반응을 통한 canonical transient receptor potential(TRPC) mRNA 발현으로 분석하였다. LPC는 세포 안의 Ca2+ 저장고에서 Ca2+ 유리와 store-operated channel(SOC) 를 통한 세포 밖의 Ca2+ 유입을 촉진시켜 세포내 Ca2+ 농도를 증가시키고, 동시에 활성산소를 생성하여 세포생존율을 저하시켰다. LPC를 처리한 HUVECs에서는 TRPC1 및 TRPC3 mRNA 발현이 유의하게(p < 0.05) 증가되었다. Chlorogenic acid는 LPC에 의해 유도된 SOC를 통한 Ca2+ 유입과 활성산소 생성을 차단하였다. 또한 chlorogenic acid는 LPC에 의해 감소된 세포생존율을 회복시켰으며, TRPC1 및 TRPC3 mRNA 발현도 유의하게(p < 0.05) 억제하였다.
이상의 결과로 보아 chlorogenic acid는 산화형 저밀도 지단백의 주성분인 LPC에 의한 TRPC mRNA 발현 증가를 약화시킴으로써 SOC를 통한 Ca2+의 유입과 활성산소 생성을 차단하여 HUVECs의 세포손상을 억제하였으며, 동맥경화증 발생을 억제하는데 도움이 될 것으로 사료된다.
Atherosclerosis is a chronic and multi-factorial inflammatory vascular disease, and reactive oxidative species (ROS) and oxidized low-density lipoprotein (ox-LDL) play a major role in its pathogenesis. Lysophosphatidylcholine (LPC) is a major phospholipid component of ox-LDL and is implicated as a critical factor in the atherogenic activity of ox-LDL. This study investigated the effects of LPC on cell viability and intracellular calcium homeostasis and the protective mechanisms of chlorogenic acid in LPC-treated human umbilical vein endothelial cells (HUVECs).
The changes in intracellular calcium concentration ([Ca2+]i) and ROS generation were measured using fura-2 acetoxymethyl (fura-2/AM) and 2',7'-dichlorofluorescein diacetate (H2DCFDA), respectively. Canonical transient receptor potential Ca2+ (TRPC) channel mRNA expressions were analyzed by reverse transcription-polymerase chain reaction (RT-PCR).
LPC increased [Ca2+]i due to both Ca2+ release from intracellular stores and Ca2+ influx through store-operated channel (SOC). LPC increased ROS generation and decreased cell viability. TRPC1 and TRPC3 mRNA expressions increased significantly (p < 0.05) after treatment by LPC. Chlorogenic acid inhibited LPC-induced Ca2+ influx and ROS generation, and restored cell viability. Chlorogenic acid significantly (p < 0.05) attenuated the LPC-induced TRPC1 and TRPC3 mRNA expressions as potent SOC blockers.
These results suggest that chlorogenic acid inhibits SOC-mediated Ca2+ influx and ROS generation via an attenuation of TRPC mRNA expressions in LPC-treated HUVECs. Therefore chlorogenic acid may be useful in inhibiting atherogenesis in endothelial cells.
