Abstract
ASIC과 TRPV는 중추신경계 및 말초신경계에서 발현되는 통각 연관 단백질로 알려져 있다. 콜라겐 유발 관절염을 사용하여 초기, 급성기, 만성기의 시기에 따른 2형 콜라겐 항체농도, 인터루킨-6 농도, 인터루킨-17 농도, ASIC1, ASIC2, ASIC3, TRPV1과 TRPV4의 발현과 약물치료에 따른 발현정도의 변화를 알아보았다. 실험군에서 대조군에 비해 2형 콜라겐 항체농도, 인터루킨-6 농도, 인터루킨-17 농도가 증가하였다. ASIC1, ASIC2, ASIC3, TRPV1과 TRPV4 모두 실험군에서 대조군에 비해 증가하였다. ASIC1, ASIC2, ASIC3은 만성기군에서 발현이 증가하였으며, TRPV1과 TRPV4는 초기군에서 발현이 증가하였다. 한편, 치료제 투여 후 ASIC1, ASIC2, ASIC3은 급성기군에서 발현이 증가하였고 초기군과 만성기군에서는 변화가 없었으며 치료기간에 따른 차이는 없었다. 치료제 투여 후 TRPV1은 급성기군에서 발현이 감소하였고 TRPV4는 급성기군에서 발현의 증가를 보였으며 치료기간에 따른 차이는 없었다. 따라서 ASIC1, ASIC2, ASIC3과 TRPV1, TRPV4가 염증성 관절염인 콜라겐 유발 관절염의 통증의 기전에 관여하는 것으로 생각된다. Different acid-sensing ion channel (ASIC) isoforms and transient receptor potential vanilloid (TRPV) have been identified and proved to be critical for development of secondary hyperalgesia in peripheral sensory neurons innervating skin and muscle. This study was aimed to investigate the ASIC and TRPV expressions in murine collagen induced arthritis (CIA). Mice were divided into 4 groups; control, early CIA, active CIA and chronic CIA. Measurement of type II collagen specific antibodies, IL-17, and IL-6 were performed. The concentrations of anti-type II collagen antibody and IL-17 in CIA were significantly higher than those in control. Expressions of ASIC1, ASIC2, ASIC3, TRPV1 and TRPV4 were markedly increased compared with those in control. ASIC1, ASIC2, ASIC3 in chronic CIA were increased compared with those in control and were increased in acute CIA treated with NSAIDs (MobicⓇ) and TNF-? blocker (RemicadeⓇ). TRPV1 and TRPV4 in early CIA were increased compared with those in control. Taken together, these results implicate that increased ASIC1, ASIC2, ASIC3, TRPV1 and TRPV are involved in the joint pain of CIA.
