Effects of endoplasmic reticulum stress on plasminogen activator inhibitor type 1 expression in rat aortic smooth muscle cells

Abstract
Endoplasmic reticulum (ER) stress is associated with numerous diseases, including Alzheimer’s disease, diabetes and cardiovascular disease. Elevated levels of plasminogen activator inhibitor-1 (PAI-1) are considered a risk factor for cardiovascular complications. However, little is known about the effect of ER stress on PAI-1 expression in vascular smooth muscle cells (VSMCs). In this study, we examined changes in PAI-1 expression induced by ER stress in cultured rat aortic smooth muscle cells (VSMCs).
Treatment of VSMCs with the ER stress inducer, tunicamycin, stimulated ATF6 protein expression, which are markers of ER stress. To investigate whether ER stress induces PAI-1 expression, the effect of tunicamycin on PAI-1 expression was evaluated by Northern and Western blot analysis. The results confirmed that tunicamycin stimulated PAI-1 mRNA and protein expression. Transient transfection assays using a PAI-1 promoter reporter construct revealed that tunicamycin increased PAI-1 promoter activity in a dose-dependent manner. Because PAI-1 expression is regulated by the transforming growth factor (TGF)-β signaling pathway, we examined whether induction of ER stress increased phosphorylation of Smad3, a downstream target of TGF-β. Indeed, Western blot analysis revealed that tunicamycin increased phosphorylation of Smad3. Mitogen-activated protein (MAP) kinases have also been implicated in PAI-1 expression; therefore, the activation of ERK and JNK MAPK was also examined. The induction of ER stress by tunicamycin increased phosphorylation of ERK and JNK, and tunicamycin-increased PAI-1 expression was blocked by ERK and JNK inhibitors.
In conclusion, this study shows that ER stress induces PAI-1 expression in VSMCs through the activation of Smad3, ERK and JNK. These data suggest that ER stress plays an important role in the development of cardiovascular diseases.
소포체 스트레스는 생리적 혹은 병리적 환경에 의해 소포체가 처리할 수 있는 능력 이상의 미성숙 단백질이 소포체 내로 유입이 되거나 소포체 내 칼슘이 고갈되면 소포체 기능에 장애가 발생하는데 이러한 상태를 소포체 스트레스 (endoplasmic reticulum stress) 라고 한다. 이러한 소포체 스트레스는 당뇨병, 심장혈관계 질환 등의 관련이 있다. 그리고 plasminogen activator inhibitor-1 (PAI-1) 은 심장혈관질환의 위험 인자로 알려져 있다. 본 실험에서는 배양된 쥐의 혈관평활근세포에서 소포체스트레스 유발이 PAI-1 발현에 영향을 미치는지를 알아보았다.
소포체스트레스를 일으키는 약제인 tunicamycin을 배양된 쥐의 혈관평활근세포에 처리한 결과 ER stress의 증가를 표시하는 ATF6의 발현이 증가되었다. Tunicamycin 처리에 의한 PAI-1의 발현이 증가하였고, PAI-1의 전사 활성도 또한 증가하였다. Tunicamycin에 의해서 Smad3의 활성도가 증가되는 것을 알아 본 결과, tunicamycin은 Smad3의 인산화와 SMAD-DNA 결합 정도를 증가 시켰다. 또한, tunicamycin의 처리에 의해서 MAPK인 ERK 와 JNK의 인산화가 증가 되었고 ERK와 JNK 억제제인 PD98059와 SP600105의 처리 할 경우 tunicamycin에 의한 PAI-1 발현증가가 억제 되었다.
본 연구를 통하여 혈관평활근세포에 소포체스트레스를 증가 시킬 경우 PAI-1 발현이 증가하고 소포체스트레스에 의한 PAI-1 발현의 증가에는 Smad3와 ERK 및 JNK 활성화가 관여함을 알 수 있었다. 따라서, 혈관평활근세포에서 소포체스트레스의 증가가 심장혈관질환의 발생에 중요한 역할을 할 수 있을 것으로 사료 된다.
