The effects of the melatonin on UVB irradiated cultured HaCaT cells

Abstract
Melatonin, the chief secretory product of the pineal gland with multiple functions in humans, was recently found to be a free radical scavenger and antioxidant. In both in vitro and in vivo experiments, melatonin has been shown to have photoprotection properties. Previously, numerous studies have been performed to assess the influence of UV light on radical formation in different cell systems or skin homogenates. But there has been little progress toward identifying the specific mechanisms of its action, especially keratinocyte. To clarify the role of melatonin as a free radical scavenger, in response to ultraviolet-B (UVB) irradiation, we investigated the effects of UVB and melatonin on cytotoxicity, lipid peroxidation, and alteration of cell cycle in cultured HaCaT cell. Cell survival curves after UVB irradiation showed dose dependent decrement pattern by trypan blue exclusion assay. Only 47% of HaCaT cells were survived at 200 mJ/cm2 UVB irradiation. The damage was associated with cell membrane lipid peroxidation, as shown by accumulation of malondialdehyde (MDA). By treatment with melatonin (10^(-7) and 10^(-9) M), a significant preventive effect was noted on the increase in the absolute number of surviving cells and the levels of MDA were markedly decreased. Quantitative analysis of DNA content of HaCaT cells was evaluated by flow cytometric analysis performed after vital staining with propidium iodide. UVB suppresses the G1 progression induced pre-G1 arrest leading to apoptotic changes of HaCaT cells and those are blocked by melatonin treatment. Taken together, our data suggest that melatonin treatment inhibits UVB-induced apoptosis of HaCaT cells, and its photoprotective effect may account, at least in part, for it’s ability to reduce lipid peroxidation. 자외선 B는 reactive oxygen species (ROS)와 활성 산소기를 생산하고 이에 대한 자연적 방어에 중요한 역할을 담당하는 피부 항산화제를 소실시킴으로써 세포 고사를 포함한 다양한 피부 손상을 유발하여 궁극적으로 피부 질환을 일으키게 된다. 멜라토닌은 송과선(pineal gland)에서 분비되는 호르몬으로, 다양한 기능을 가지는데, 특히 유리기 제거제 기능 및 항산화 기능이 알려져 있으며, 이런 기능으로 자외선에 의한 피부 손상을 막아주는 역할을 한다. 최근 국내외적으로 다양한 세포군을 대상으로 자외선에 의한 광생물학적 변화와 멜라토닌의 효과에 관한 임상적 기초과학적 연구가 활발히 진행되고 있고, 피부 각질형성세포를 대상으로 한 연구도 소수 있었으나, 세포 고사를 포함한 세포 손상에 멜라토닌이 미치는 영향에 대한 자세한 연구는 없었다. 따라서, 이 연구는 자외선-B에 의하여 발생하는 각질형성세포 손상에 대한 멜라토닌의 효과를 알아보고자, 인간 각질형성세포주인 HaCaT 세포를 배양하고 자외선-B를 조사한 후 멜라토닌을 다양한 농도로 처리 하여 세포 독성과 세포막 지질의 과산화 작용, 세포 주기 변화 등을 관찰하여 다음과 같은 결과를 얻었다.
자외선-B를 조사한 결과 세포 생존율은 자외선 조사양과 반비례하였으며 200 mJ/cm²의 자외선-B를 조사한 결과 그 생존율은 약 47%였고, 세포막 지질의 산화 손상 정도를 나타내는 malondialdehyde (MDA)는 자외선 조사량에 비례하여 증가하였다. 이러한 세포사는 MDA의 수치가 증가된 것으로 보아 세포막의 지질과산화작용과 연관이 있음을 알 수 있다. 멜라토닌 10^(-9) 과 10^(-7) mol 처치 시에 HaCaT 세포의 생존율이 증가되었고 그와 동반하여 MDA 수치도 감소함을 알 수 있었다. 또한 10^(-9) 과 10^(-7) mol 멜라토닌 처치 시 자외선-B에 의한 HaCaT 세포의 apoptotic 변화도 현저하게 떨어뜨렸다. Propidium iodide (PI) 염색 후 flow cytometry 분석법으로 알아본 세포 주기 변화는 자외선-B에 조사 후 pre-G1 apoptotic population이 비조사군에 비해 증가되는 세포 고사 형태를 보였으나, 10^(-9) 과 10^(-7) mol의 멜라토닌을 처치한 세포군 에서는 pre-G1 apoptotic population이 감소되어 멜라토닌이 apoptosis에도 영향을 줌을 알 수 있었다.
이상의 결과로 배양된 HaCaT세포에서 자외선-B 조사는 apoptosis에 의한 세포사를 유도하며 이러한 세포사는 세포막의 지질과산화작용과 연관이 있음을 알 수 있었다. 멜라토닌 처치는 자외선-B 조사에 의한 apoptosis를 억제하며 세포막의 지질과산화작용을 억제함으로써 자외선-B에 대한 보호 작용을 나타낼 것으로 생각된다.
