Abstract
After injection of 0.1ml compound 48/80(1mg/ml) into footpat of rat, the morphological changes of mast cell, its disrupted granules, and relations between mast cells and adjacent cells in the popliteal lymph nodes were observed by light and electron microscopes. Results were as followings: Light microscopic observations: Mast cells in the lymphatic sinus were degranulatd in maximum at 2 hour after injection of compound 48/80. Most of degranulated granules were observed in the lymphatic sinus and within some cells supposed to be macrophages and reticular cells. Mast cells in the lymph nodule had a few of granules. Elecron microscopic observations: Mast cells in the lymphatic sinus at 2 hour after injection of compound 48/80 were degranulated severely and their disrupted granules were phagocytosed by macrophages and reticular cells, or existed in free form in the lymphatic sinus. Macrophages which were not contacted with mast cell phagocytosed a few of granuls, while reticular cells phagocytosed a number of granules. Pattern of interdigitation through microvilli-like cytopasmic processes between mast cells and reticular cells in experimental group was more complicated than that in normal group. The lymphatic sinuses at 2 hour after injection of compound 48/80 were dilated and contained with eosinophils, neutrophils, and plasma cells. These cells had no mast cell granules in their cytoplasm. Plasma cells with dilated cisternae of rough endoplasimc reticulum were also found in the lymphatic sinus occasionally. Mast cells of the lymph nodule in control group contained with fewer granles than those of the lymphatic sinus. Many vacuoles in cytoplasm of mast cell were observed among the electronlucent granules after injection of compound 48/80. These mast cells were contacted with reticular cells in a similar pattern to those of lymphatic sinus, but their microvilli-like processes wererather simple. The cistrnae of rough endoplasmic reticulum of some plasma cell in these region were dilated and contained with electron dense granules. From the above results, it was suggested that the response of compound 48/80 to mast cells in the lymphatic sinus was different from that in the lymph nodule, and that reticular cells were concerned with secretion of mast cells rather than macrophages.
