Abstract
Sterol regulatory element-binding proteins (SREBPs) are involved in
fatty acid synthesis and transcription factors regulating cellular lipid and
cholesterol synthesis. SREBP-1c is mainly expressed in liver, muscle
and adipose tissue and regulates gene expressions of glucose and lipid
metabolic enzymes.
Lipocalin (LCN)-2 belongs to the family of lipocalin protein and is
expressed in multiple tissues and organs as an acute-phase protein.
According to recent studies, LCN-2 is known as a critical protein in
obesity, insulin resistance and hepatitis. However, there is no complete
understanding about the molecular mechanism of LCN-2 gene regulation
by SREBP-1c.
In this study, a mouse model of alcoholic fatty liver disease was used to figure out transcriptional regulation of LCN-2 gene expression.
Eight-week old male mice were divided into two groups: wild-type
(WT) and SREBP-1c knock-out (KO) mice (n = 6 per genotype).
Alcohol was provided at 5% of the Lieber-DeCarli liquid diet for 3
weeks to induce hepatic steatosis. As a result, LCN-2 expression was
decreased in SREBP-1c KO mice compared to WT. In addition, serum
biomarkers such as aspartate amino transferase (AST) and alanine
amino transferase (ALT) were lower in SREBP-1c KO mice compared
with WT mice. Expression of pro-inflammatory cytokines; TNF-α and
MCP-1, which are induced by liver disease, were also less in SREBP-1c
KO mice. From ChIP-seq, we confirmed LCN-2 gene contains
consensus sequences for binding SREBP-1c. ChIP assay showed that
binding site of SREBP-1c is located on the LCN-2 promoter. Taken
together, this study suggests that SREBP-1c regulates gene expression
of LCN-2 and could be a possible mechanism to repress alcoholic fatty
liver disease such as hepatic steatosis, hepatitis and cirrhosis, which
might be used for the preventive strategies in treating chronic liver diseases caused by alcohol-induced imbalance of lipid metabolism.
지방산 합성에 관하여 스테롤조절인자 결합 단백질(sterol regulatory
element-binding protein, SREBP)은 세포내 지방과 콜레스테롤 대사를 조
절하는 전사인자로 잘 알려져 있다. SREBP-1c는 간, 근육 및 지방에 많이
발현되며, 포도당 및 지방대사에 관여하는 효소들의 유전자발현을 조절하는
것으로 알려져 있다. 리포칼린-2(Lipocalin-2)는 리포칼린 계열의 단백질에
속하며 염증에 급성으로 반응하는 단백질로서 여러 장기 및 기관에서 발현
이 된다. 최근 연구에 따르면 리포칼린-2는 비만, 인슐린 저항성 및 염증간
등에 중요성을 띠는 단백질로 알려져 있다. 그러나 SREBP-1c에 의한 리포
칼린-2의 발현 조절에 대한 기전 연구는 전혀 알려진 바가 없다. 알콜성
간질환 모델에서 SREBP-1c에 의한 리포칼린-2의 발현 조절을 규명하기 위
해서 8 주령의 정상 및 SREBP-1c 결핍 마우스를 3 주 동안 Lieber-Decarli
식이를 이용하여 에탄올을 투여한 후 간질환 형성을 유도한 결과, SREBP-1c 결핍 마우스가 정상 마우스에 비해 리포칼린-2의 발현이 감소함을 확인하였
다. 간염 수치의 혈청 지표인자인 aspartate amino transferase 및 alanine
amino transferase도 SREBP-1c 결핍 마우스에서 감소되었다. 간질환이 유
도되었을 때 증가되는 염증성 사이토카인인 TNFa와 MCP-1의 발현이
SREBP-1c 결핍마우스에서 감소하였다. 크로마틴 면역침강법 염기서열 분
석을 통해 SREBP-1c에 의해 리포칼린-2에 결합하는 서열을 확인하였고,
크로마틴 면역침강법을 통해서 리포칼린-2의 프로모터에 SREBP-1c가 결
합할 수 있는 부위가 있음을 검증하였다. 결론적으로 이번 연구에서 리포칼
린-2는 SREBP-1c에 의해 발현을 조절하여 알콜성 간질환에 의해 유발되
는 지방간, 간염 및 간경변 등을 저해 할 수 있는 새로운 기전으로 규명하
였으며, 만성적인 에탄올 섭취에 의한 지방대사 불균형이 가져오는 각종 염
증성 간질환에 대한 예방적 전략을 마련할 수 있을 것이다.
