오미자 부산물 발효액을 활용한 피부장벽 및 피부주름 개선 효과

Abstract
Schisandra chinensis is known for anti-bacteria, anti-clotting activities. Schisandra chinensis is mainly processed as a tea, marmalade, beverage products, and 40% of by-products is produced from pulp, seed and shell. However, a large quantity of its by-product is discarded during the processing. In this study, to prove the possibility of developing various materials using by-product, fermented Lactobacillus paracasei Schisandra chinensis extract (LPSC-E) was treated to human skin cells and artificial skin. Moreover, the skin damage and wrinkle formation were confirmed by UVB induction. This study was aimed to investigate the anti-skin barrier and anti-wrinkle effects and mechanisms of LPSC-E. The exposure of UVB at 300 mJ/cm2 for 5 h into HaCaT keratinocytes decreased protein and mRNA expressions of involucrin and filaggrin skin barrier factors to 66% and 64%, respectively. However, when the skin cells treated with 100 μg/mL of LPSC-E, protein and mRNA expressions of involucrin and filaggrin were significantly decreased to 84% and 79% compared with non-treated cells. LPSC-E selectively inhibited the UVB-induced ERK-1/2 and c-FOS expression in HaCaT cells. In an artificial skin, treatment of UVB at 80 mJ/cm2 for 24 h induced significant damage to the epidermal layer along with decrease in the mRNA expression of involucrin and filaggrin. Upon treatment of 100 and 200 μg/mL LPSC-E, artificial skin showed higher recovery than the positive control (WY14643 50 μM). Furthermore, HDF (Human Dermal Fibroblasts) cells treated with 60 mJ/cm2 of UVB for 4 h significantly decreased mRNA and protein expression of type I collagen. However, when these cells treated with 100 μg/mL of LPSC-E both mRNA and protein expression were up-regulated. Treatment with LPSC-E at 100 μg/mL considerably up-regulated procollagen expression. These results demonstrate that LPSC-E has anti-skin barrier and anti-wrinkle effects on skin cells and artificial skin models through modulation of the expression levels of involucrin, fillaggrin, and type 1 collagen, which may suggest that LPSC-E may be applied as a potential cosmetic ingredient for the development of anti-skin barrier and anti-wrinkle cosmetic products.
오미자(Schisandra chinensis)는 항균 및 항 응고 등의 생리활성 효능을 가짐이 알려져 있고, 차, 청, 음료 등으로 제조되면서 과육, 씨, 껍질 등 40% 수준의 부산물이 발생하나, 아직까지 오미자 부산물의 산업적 활용도를 찾지 못해 대부분이 폐기되고 있는 실정이다. 이에 본 연구에서는, 오미자 부산물을 활용한 다양한 소재 개발의 가능성을 증명하기 위하여, 오미자 부산물의 발효액을 제조하여 인간 피부세포 및 인공피부에 처리한 후, UVB에 의해 유도되는 피부장벽 손상 및 주름 형성에 대한 억제 효능들을 확인하였다. 먼저, 오미자 부산물에 균주 Lactobacillus paracasei를 혼합하여 30 ˚C에서 48 시간 발효한 후 30% 에탄올로 추출함으로써 발효액(LPSC-E)를 제조하였다. 이어, 각질세포(HaCaT)에 UVB를 300 mJ/cm2의 양으로 5 시간 조사하면, 피부장벽 유지인자 involucrin과 filaggrin의 mRNA 및 단백질 발현 모두가 각각 66%와 64% 감소함을 확인하였다. 이와 반대로, 100 μg/mL LPSC-E를 처리 시, 비 처리 대조군에 비해 발현이 각각 84% 및 79% 회복됨을 확인하였다. 또한, 동일 조건에서 성장인자 ERK-1/2 및 c-Fos의 발현도 부분적으로 회복되었다. 한편, 인공피부에 80 mJ/cm2의 UVB를 24 시간 조사하면, 표피층이 손상되면서 involucrin과 filaggrin의 mRNA 발현이 감소되었으나, 100 또는 200 μg/mL의 LPSC-E를 처리하면 발현 감소가 상쇄되며, 특히 200 μg/mL 처리 시 양성 대조군(WY14643) 보다 높은 복원 효능을 나타내었다. 또한, 진피섬유아세포(HDF)에 60 mJ/cm2의 UVB를 4 시간 조사하면, type 1 collagen의 mRNA 및 단백질 발현이 감소하고, procollagen의 발현도 67% 감소하였다. 역으로, 100 μg/mL의 LPSC-E를 처리하면, type 1 collagen의 발현 감소가 상쇄되며, procollagen의 합성을 촉진함을 증명하였다. 이러한 결과들은, 오미자 부산물이 피부장벽 손상 개선(보호)제 및 주름 개선용 화장품 개발의 원료로서 범용성을 가질 수 있음을 제시해 준다.
