Abstract
Cyclins are prime cell cycle regulators and are central to the control of major check point in eukaryotic cells. But, the structural change and overexpression of cyclin mRNA demonstrated to association of oncogenesis in some cancers. Cyclins were first identified in marine invertebrates on the basis of their dramatic cell cycle periodicity during meiotic division. Cyclins function by forming a complex with and activating a family of cyclin-dependent protein kinase(CDKs), at various stages in the cell cycle. They fall into three categories ; A-type, B-type, and G1cyclins(cyclins C, D1-D3, and E). In this study, we have examined expression of cyclins(A, D1 and E) messenger RNA in basal cell carcinoma and squamous cell carcinoma(SCC) tissues and SCC cell lines by semi-quantitative RT-PCR method, immunoperoxidase stain and the results were as follows: 1. Expression of cyclin A, D1 and E genes in basal cell carcinoma, squamous cell carcinoma and SCC cell line were more increased compared with normal skin tissues. 2. Expression of cyclin A, D1 mRNA in squamous cell carcinoma were more increased compared with basal cell carcinoma. 3. Expression of cyclin E mRNA was detected in two SCC tissue and not detected in BCC tissues. 4. Expression of cyclin A, D1 and E mRNA in SCC cell line remarkably increased compared with basal cell and squamous cell carcinoma tissues. 5.Immunohistochemical stain of cyclin D1 revealed the increased production of the cyclin D in SCC tissues. In conclusion, these data suggest that cyclin A, D1 and E play a role in a subset of oncogenesis of skin cancer. However, further studies including gene analysis and Southern analysis are needed to clarify the relation between cyclins, oncogenes and other factors in oncogenesis of human skin cancers.
