간세포암종과 MMP1 유전자 단일염기 다형성의 관련성

Abstract

간세포암종은 세계적으로 흔한 악성종양 중의 하나로 주된 사망 요인 중의 하나이다. 간세포암종의 발병원인 중 유전적인 원인도 매우 중요한 인자이다. Matrix metalloproteinase 1 (MMP 1)은 기저막과 세포외 기질의 주성분을 분해하는 단백 분해효소로서 암을 비롯한 다양한 질병의 발생과 전이에 관여한다. 본 연구에서는 한국인 간세포암종 환자에서 MMP1 유전자 프로모터 위치 -1607, -519의 다형성에 대하여 조사하였으며 이러한 소견들과 간세포암종의 발생과 임상 양상과의 관련성을 조사하였다. 50세 이상의 간세포암종으로 진단 받은 남자 환자 124명과 건강한 대조군 147명의 혈액에서 pyrosequencing method를 이용하여 유전자형을 분석하였고 간세포암종 발생과의 상관관계, 임상 양상 등에 대하여 조사하였다. MMP1 유전자의 -1607 위치에서의 염기다형성이 간세포암종의 발생과 유의성이 있는 것으로 나타났다. 일배체형 2GA를 하나라도 가지고 있는 일배체형이 없는 경우에 비하여 간세포암종의 발생이 2.34배 높은 것으로 나타났다. 간세포암종 환자에서 MMP1 유전자 -1607, -519 위치에서의 단일 염기 다형성과 간세포암종의 임상적인 연관성은 없는 것으로 나타났다. 50세 이상의 남자 한국인 간세포암종 환자에서 MMP1 유전자 -1607 위치와 2G 대립유전자가 간세포암종의 발생과 관련이 있을 것으로 추정된다.

Hepatocellular carcinoma (HCC) is common and main cause of death in Korean. Genetic factors are also important contributing factors to the development of HCC. Matrix Metalloproteinase 1 (MMP1) is the primary collagenase elaborated by fibroblasts and monocyte. It cleaves substrates at a fibrillar triple helix at a specific site, producing fragments that can be further degrades by other MMPs. We examined whether single nucleotide polymorphisms (SNPs) of the MMP1 gene are associated with HCC in Korean population. I genotyped three SNPs in the MMP1 gene using the pyrosequencing method in 124 subjects with HCC and 147 normal subjects over 50 ages. I evaluated the association of SNPs of MMP1 gene and clinical characteristics such as tumor type, portal vein thrombosis, tumor invasion, tumor stage, tumor size, and metastasis. One SNP (-1607) of MMP1 gene were found to be associated with HCC in recessive model after adjusted age. In haplotype analysis, one haplotype (2GA) was detected to be associated with HCC in recessive model (OR: 2.34, CI: 1.07-5.08) There was not found to be association between genotype distribution of MMP1 gene at -1607 and -519 position and clinical characteristics of HCC patients. These results suggest that the MMP1 gene -1607 2G contributes to genetic susceptibility to HCC in Korean population.
