Abstract
A study was made on the changes in activities of the followings during 6 post-operative days : Plasma membrane, microsomal, ,nuclear and cytosolic 5'nucleotidase and gamma-glutamy1 transpeptidase of regenerating rat livers, ant rat serum 5'nucleotidase and gamma-glutamy1 transpeptidase after 70% (median and left lateral lobes) partial hepatectomy. The activities of alanine aminotransferase in both serum and cytosol of regenerating rat liver were also measured. The activities of 5'-nucleotidase in the normal rats hepatic subcellular fractions were " 59.29±11.35(n mol Pi/mg protein/min)in the plasma membrane ; 52.86±3.88 in the microsome ; 10.56±2.12 in the mitochondria ;11.48±3.62 in the nuclei, and 3.55±0.54 in the cytosol. Those of gamma-glutamy1 transpeptidase were: 1.96±0.46 (n mol p-nitroaniline/mg protein/min) in the plasma membrane ; 3.38±0.62 in the microsome ; 0.88±0.12 in the mitochondria ; 1.40±0.13 in the nuclei, and 0.76±0.12 in the cytosol. Normal activity of alanine aminotransferase was 330±56 Karmen unit/mg protien in the rat hepatic cytosol. After partial hepatectomy in the rats, activities of plasma membrane bound gamma-glutamy1 transpeptidase in regenerating livers significantly increased in the span between the second and the third days but plasma membrane bound 5'-nucleotidase showed no change. The activity of microsomal 5'- nucleotiadase in the regenerating liver tremendously from the second to the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 transpeptidase also elevated from the second and the third days after partial hepatectomy. Tomy activity of mitochondrial 5'-nucleotidase in the regenerating liver decreased between the third and the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 trsnspeptidase also elevated from the second and the third days after partial hepatectomy. The activity of mitochondrial 5'-nucreotidase in the regenerating liver decreased between the third and the sixth days of partial hepatectomy. The hepatic nuclear and cytosolic 5'-nucleotidase decreased in the regenerating stage. The decreased values were seem after two days in cytosolic 5'-nucleotidase and after three days in nuclear 5'-nucleotidase following the operations. However, no significant changes in hepatic mitochondrial, nuclear and cytosolic gramma-glutamy1 were ntoed through out the experiments. The activity of serum alanine aminotransferase markedly elevated from 12 hours to three days after partial hepatectomy, but serum 5'-nucleotidase and gamma-glutamy1 transpeptidase activities remained normal.
