- Abstract
- Type Ⅶ collagen is the predominant component of the anchoring fibrils and both dermal fibroblasts and epidermal keratinocytes in culture synthesize type Ⅶ collagen. The presence of anchoring fibrils is apparently critical for the stability of the cutaneous basement membrane zone, because abnormalities in quantity of quality of anchoring fibrils manifest as dystrophic forms of epidermolysis bullosa(EB). But there is little known about effects of cytokines on type Ⅶ collagen gene expression. We have examined the response of the fibroblasts and keratinocytes in type Ⅶ collagen gene expression to transforming growth factor-β (TGF-β) as well as tumor necrosis factor-α (TNF-α), which have been shown to modulate the expression of the extracellular matrix genes using Northern blot hybridizations ···
