임상 검체에서 PCR을 이용한 수두-대상포진바이러스 DNA의 검색

Abstract

The polymerase chain reaction (PCR) was used to detect varicella-zoster virus (VZV) DNA in the herpes zoster vesicle specimens and paraffin section specimens from patients. Oligonucleotide pairs for 44 kilodalton protein gene were used as primers and VZV Oka strain was used as a con¬trol. Positive results were obtained in 10 of 10 vesicle specimens and 6 10 paraffin section speci¬mens. Detection of PCR products by direct agarose gel electrophoresis (EP) showed positive results in 10 of 10 vesicle specimens and only 3 of 10 paraffin sections. But with southern blot hybri¬dization, we could obtain positive results in 10 of 10 vesicles and 6 of 10 paraffin sections. Slot blot hybridization data also showed the same results as in the case of southern blot analysis. Dig- oxigenin labeling and detection method was used in southern and slot blot hybridiations in order to obtain data rapidly and more conveniently. In conclusion, these results showed that the PCR provides a useful tool for the early diagnosis of VZV infections and the detection rate of PCR products is increased by agarose gel EP with south¬ern or slot blot hybridization.
